Regulatory Status

Who Should Not Use Enclomiphene?

Warnings and Precautions

1. Blood Clot Risk (Thromboembolic Events)As a selective estrogen receptor modulator (SERM), enclomiphene may be associated with an increased risk of blood clots, including deep vein thrombosis (DVT) and pulmonary embolism (PE). This risk may be higher in individuals with pre-existing risk factors such as obesity, smoking, prolonged immobility, or personal/family history of clotting disorders. Seek immediate medical attention if you experience leg pain, swelling, warmth, or redness, or if you develop sudden shortness of breath or chest pain.

2. Estrogen EffectsAlthough enclomiphene is primarily an estrogen antagonist, changes in estrogen balance may occur. Some men may experience elevated estradiol levels. Your healthcare provider should monitor hormone levels during treatment and adjust therapy as needed.

3. Liver FunctionWhile enclomiphene has not been directly linked to liver toxicity in clinical studies, SERMs as a class require caution in patients with liver conditions. Inform your provider if you have any history of liver disease.

4. Vision ChangesVisual disturbances have been reported with clomiphene citrate (the parent compound) and may potentially occur with enclomiphene. Report any changes in vision, including blurred vision, spots, flashes of light, or difficulty with night vision, to your healthcare provider immediately.

5. Mood ChangesSome individuals may experience mood changes, including depression, irritability, or mood swings. However, studies suggest these effects are less common with enclomiphene than with clomiphene citrate. Report significant mood changes to your provider.

6. Hormone MonitoringRegular blood tests are recommended to monitor testosterone, LH, FSH, estradiol, and hematocrit levels. Your provider will use these results to assess treatment effectiveness and safety.

Common Side Effects

Enclomiphene is generally well-tolerated. Side effects reported in clinical trials were mostly mild and included:

  • Headache (approximately 3% of patients)
  • Hot flashes
  • Dizziness (approximately 1% of patients)
  • Nausea
  • Abdominal discomfort
  • Fatigue
  • Mood changes
  • Acne

Most side effects are transient and tend to diminish as the body adjusts to treatment.

Less Common Side Effects

The following have been observed less frequently:
